The PathHunter ACVRL1/BMPR2 Dimerization eXpress kit provides an easy-to-use cell based assay to study drug candidates (small molecules or biologics) targeting receptor-receptor interaction. This kit assesses ligand (e.g. GDF2/BMP9) induced dimerization of ACVRL1/BMPR2 receptors. This product uses EFC technology. The cell line included overexpresses ACVRL1 target protein fused to PK alongside BMPR2 target protein fused to EA. Upon ligand-induced dimerization of ACVRL1/BMPR2, detectable EFC signal is produced. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. The PathHunter GPR35 Activated GPCR internalization eXpress kit provides an easy to-use cell based assay to study drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This kit uses EFC technology to measure ligand-induced internalization of GPR35 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged GPR35. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting GPR35-arrestin complex is internalized to the early endosome, leading to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Cayman Chemical Steatosis Colorimetric Assay
Steatosis, also known as fatty liver, is a pathological process characterized by abnormal accumulation of lipids within cells. Cayman’s Steatosis Colorimetric Assay provides a convenient tool for evaluating the steatosis risk of drug candidates. In this assay, Oil Red O is used to stain neutral lipids in hepatocytes. Lipid accumulation is then quantified using a plate reader after the dye is extracted from the lipid droplets. Chloroquine is included in the kit as a positive control.
Cayman Chemical Acid Phosphatase Colorimetric
Cayman’s Acid Phosphatase Assay provides a convenient method for detecting AP activity in plasma, serum, urine, and semen. The assay utilizes para-nitrophenyl phosphate (pNPP) as a chromogenic substrate for the enzyme. In the first step, AP dephosphorylates pNPP. In the second step, the phenolic OH-group is deprotonated under alkaline conditions resulting in p-nitrophenolate that yields an intense yellow color which can be measured at 405-414 nm. The kit provides all reagents needed to assay AP activity, and includes L-tartrate, an inhibitor of non-tartrate resistant APs.
