This kit assesses ligand (e.g. secretin) based activation of SCTR GPCR activity via detection of β-arrestin recruitment. The included cell line overexpresses ProLink-tagged SCTR and EA-tagged β-Arrestin-2. Activation of SCTR stimulates the recruitment of β-Arrestin-2 and produces EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. The PathHunter CMKLR1 Activated GPCR internalization eXpress kit provides an easy to-use cell based assay to study drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This kit uses EFC technology to measure ligand-induced internalization of CMKLR1 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged CMKLR1. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting CMKLR1-arrestin complex is internalized to the early endosome, leading to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Chemotaxis, a type of cell migration, is a highly integrated, multistep process that orchestrates embryonic morphogenesis, tissue repair and regeneration. The initial response of a cell to a migration-promoting agent is to polarize and extend protrusions in the direction of the attractant; these protrusions can consist of large, broad lamellipodia or spike-like filopodia that are driven by actin polymerization and can be stabilized by extracellular matrix (ECM) adhesion or cell-cell interactions. The CytoSelect™ Cell Migration Assay Kit utilizes polycarbonate membrane inserts (8 µm pore size) to assay the migratory properties of cells. The kit contains 5x96 polycarbonate membrane inserts with 5 um pores in each of five 96-well plates, through which migratory cells are able to pass. Cells are quantified using the provided fluorometric dye. The 5 µm pore size is optimal for measuring the migration of monocytes and macrophages.
